sds-page 分子量分析怎么都不对，求帮忙 已有3人参与

我用了7%SDS，然后做western blot，就用那个照相设备出了以下图片，电脑里面是用image lab去求它的分子量，由于那个marker没有显示出来，我用了其它的方法把它标记在样品上了，关键是，分析分子量分布的时候，感觉怎么都是不对的 就好比泳道9，那个分子量怎么说都比250kda要高，可是linear上面测得它却只有240kda，这不科学呀~ 我弄了老半天也不知道是哪里错了，所以只好来求助了  60s.jpg |

I am trying to comment on your problem from a amateur's point of view. the molecular weight markers are indicating the molecular weight in a proximate manner, not at all accurate, nor are they linear. the markers from different companies behave differently. For me, fermentas and bio-rad markers are better. but you cannot expect very accurate MW indication. Usually, we are trying to get positive control loading to confirm that the band you see is what you want. good luck. |

one more comment: if you really want to predict the molecular weight in this way, you should do with a molecular weight marker with more broader spectrum. that is to say, the largest molecular weight indicator should be over 280kDa and you can still see the linear regression. In your case, the largest marker is 250kDa and your target band is out of the range. You cannot do linear regression outside your standard since you never know if that part is still linear. |
