研究领域:用于诊断和癌症研究的先进单细胞技术

细胞包含多层次的信息,从遗传学到化学成分、物理特性,最重要的是生物功能。这种个体异质性已成为临床诊断和疾病研究的主要挑战。为了解决这个问题,我们一直在开发先进的单细胞技术,用于细胞分类和分析。与合作者一起,我们在癌症研究、细菌耐药性诊断和药物筛选方面开发了这些工具。


以下是我们希望围绕这一研究主题进一步发展的项目。


1) 纳米尺度热与机械生物学。温度是许多细胞功能的关键。然而,通过我们最近开发的水扫描热显微术,以纳米级分辨率对细胞进行直接热测量已经成为可能。这个项目提供了一个令人兴奋的机会,进行世界领先的研究,将利用这一创新技术来测量局部温度和机械性能的细胞。这些基本的测量将解决生物学以及纳米颗粒介导的癌症热治疗中尚未解决的问题。


2) 高通量拉曼活化细胞分选。拉曼活化细胞分选(RACS)已成为一种强有力的工具,可以“深度挖掘”生物系统,揭示其异质性,并在细胞功能和基因组成之间建立联系。本计画将以我们首创的流动式持续RACS平台为基础,发展一套结合深度学习的高通量系统,用于癌症的早期诊断及治疗药物的筛选。


这些项目是跨学科的,我们欢迎积极的工程师,物理学家,生物学家和材料科学家的申请。学生将学习先进的制造方法、成像分析和其他分析科学。s/he将有很多机会与我们的合作者一起旅行和工作。

Research area:Advanced Single-cell technologies for diagnosis and cancer research

A cell contains multi-layered information, ranging from genetics to chemical composition, physical properties and most importantly, biological functions. This individual heterogeneity has become a major challenge in clinical diagnosis and studying diseases. To address this problem, we have been developing advanced single-cell technologies for both cell sorting and analysis. Together with collaborators, we have exploited these tools in cancer research, bacterial resistance diagnosis, and drug screening.

Below are projects that we’d like to further develop around this research theme.

1)Nanoscale thermal and mechanical biology. Temperature is key to many cellular functions. However, direct thermal measurements of cells with nanoscale resolution has just been made possible through our recent development of aqueous Scanning Thermal Microscopy. This project offers an exciting opportunity to undertake world leading research that will employ this innovative technology to measure local temperature and mechanical properties of cells. These fundamental measurements will address unsolved questions in biology as well as nanoparticle-mediated thermal therapy for the treatment of cancer.

2)High throughput Raman activated cell sorting. Raman-activated cell sorting (RACS) has emerged as a powerful tool for ‘deep mining’ a biological system, revealing its heterogeneity as well as establishing links between cellular function and genetic makeup. Building upon our pioneering continuous RACS platforms in flow, this project will develop high throughput system integrated with deep learning for early diagnosis of cancer and screening of therapeutics.  

These projects are cross-disciplinary and we welcome applications from motivated engineers, physicists, biologists and material scientists. The student will learn advanced manufacturing methods, imaging analysis and other analytical sciences. There will be many opportunities for s/he to travel and work alongside our collaborators.
