Title: Bio-inspired Synthesis of Aromatic Fungal Polyketides

Short description of possible research topics for a PhD:
Polyketides are an important class of highly biologically active secondary metabolites.
A lot of antibiotics or antitumor agents are derived from polyketides. Due to the antibiotic
resistance problem, the synthesis of new biologically active compounds is crucial.
Recently, a new polyketide called talaroketal A has been isolated from the fungus
Talaromyces stipitatus. This compound has been identified as a dimer of corymbiferan with a
structurally intriguing spiroketal moiety. The large number of natural products exhibiting a
corymbiferan skeleton with original structures as well as their interesting biological activities,
make this family a target of choice for the synthesis. Based on the biosynthesis of aromatic
polyketides, we have developed a synthetic strategy relying on two cyclisations of a
polyketone analogue to obtain corymbiferan D, a linchpin intermediate for our study.
The first objective will be the synthesis of monomeric natural products such as
neonectrolide A and sclerodin. Then, the dimerisation process will be studied and applied to
obtain some very complex natural products (bacillisporin A and talaroketal A). In addition,
these natural products will be biologically tested in order to find new activities. 返回小木虫查看更多
