[求助] ELISA检测蛋白相互作用。

各位大侠，救命啊。鄙人用ELISA检测蛋白质相互作用，就是测不出来。我的protocol如下：
1  Coat the microplate with the first protein at 37℃ for 3 h ;

2  Block the plate with 3% BSA at 37℃ for 2 h;

3  The second protein with  GST-tag was added to each well, incubated for 2 h at  37℃, then wash twice with PBS ;

4  Add  anti-GST Biotin conjugated (diluted 1:1000) in PBS-0.05% Tween-20, and incubated at 37℃ for 1 h, and then washed three times in PBS-0.05% Tween-20;

5   Add Streptavidin conjugated Alkaline Phosphatase in PBS-0.05% Tween-20,and incubated at 37℃ for 1 h, and then washed three times in PBS-0.05% Tween-20;

6   The color reaction was developed with p-NPP.

7  Stop the reaction and teh absorbance was read by a microplate reader at 405nm.
似乎蛋白之间的相互作用很弱，经不起折腾，有哪位大侠有成功经验或好的建议，帮帮我吧。另外，我现在只能用ELISA，其他方法条件不具备。
