请大神帮忙看看退修意见!

意见如下,我有几个小问题。一是Comments to the Author必须要回复么?而是为什么Reviewer: 1下面没有Points for Major Revision ,而Reviewer: 2下面有两个?希望有经验的大神指点迷津!(杂志是Microbial Drug Resistance,有点水,请不要嫌弃我。。。)

line 11--I would not consider K. pneumoniae an opportunistic infection.  It is a true pathogen that can affect normal hosts.  

fosfomycin spelling must be corrected throughout

You describe absolutely nothing else about the resistance phenotype in these isolates such as the presence of various beta-lactamases and FQ resistance likely on plasmids, loss of outer membrane porins,etc.  Not all of the TG resistant isolates revert their phenotypic resistance.  You should determine if they have lost a plasmid.  Whole genome sequencing is even a possibility and is not that expensive or difficult anymore.

Comments for Manuscript Reference “Contribution of efflux pumps to tigecycline resistance in carbapenem-resistant Klebsiella pneumoniae isolates”. Overall the manuscript is good and elaborate and definitely will be of great interest in microbial Drug Resistance. I therefore recommend this article for its acceptance. However, some issues are there, and these must be addressed before its acceptance.
Points for Major Revision
1.        The paper begins with a description of collection one tigecycline resistant and four induced mutant tigecycline resistant isolates. But author have not shown the clonality of isolates. To know the clonality of isolates author should do some typing assay like PFGE or MLST.
2.        In page 6, line 21, author have mentioned that mutation in acrR  or  ramR upregulate the expression of efflux pump, then how they will explain upregulation of the expression of efflux pump in the absence of acrR  or  ramR mutation in induced mutant tigecycline resistant isolates in their result.
3.        Since four induced mutant tigecycline resistant isolates were isolated from four different susceptible isolates, then why author had taken tigecycline susceptible isolate K467 as control. Author should use respective susceptible isolate as control.
Points for Major Revision
1.        Page5, Line5, remove “and so on” and complete the sentence.
2.        Arrange the number of reference started first from introduction and follow with materials & method and discussion. 返回小木虫查看更多
