Glycobiology with focus on chemoenzymatic synthesis of polysaccharides
Subject description
The subject field is within glycobiology, with a specific interest in the reconstruction of the active polysaccharide synthase complex in vitro, for the purpose of chemoenzymatic synthesis of biologically important polysaccharides. The total chemical synthesis of homogeneous polysaccharide is a formidable task. One of the major challenges is in the construction of optically pure polysaccharide by chemical synthesis in synthetically useful quantity. Chemoenzymatic synthesis of polysaccharides has high potential for large-scale synthesis for biomaterial applications and carbohydrate-based drug discovery, but this technique is largely unexplored. A major advantage of chemoenzymatic synthesis is its high regio- and stereo-selectivity. The reaction is performed in mild condition, in water at pH 7, as such it is cost-effective and environmental friendly for the production of important biopolymers. The initial focus is to optimize the expression and purification of polysaccharide synthases; with established protocols including but not limited to protein expression, purification, and functional reconstitution of polysaccharide synthase in nanodisc. Our laboratory belongs to the Division of Glycosciences at KTH, with extensive collaborations with the Wallenberg Wood Science Centre, the Australian Research Council Centre of Excellence in Plant Cell Walls and Adelaide Glycomics Centre. Duties The successful Postdoctoral candidate is expected to work independently on chemoenzymatic synthesis, and to contribute to ongoing research programs as a team member within the Division as required. Research work is to conducted at the AlbaNova University Centre, Divison of Glycoscience, School of Biotechnology, KTH https://www.biotech.kth.se/glycoscience/Main_Page with close interaction and joint research with ARC Centre of Excellence in Plant Cell Walls, and established groups from the Wallenberg Wood Science Center.
Eligibility
The eligible candidate must have a Ph.D. and have a strong background in protein expression and purification. Experience in the fields of membrane protein and enzymology are preferred but not required. Employment
Initially for one year, may be extended dependent on performance.
