16S高通量测序中的Multiple testing correction
最近投了一篇文章,审稿意见中审稿人提了这么一条:
1. Multiple testing correction should be applied to category wide tests of taxa. 除此之外没有更加具体的信息。和许多师兄讨论过都没有明白这个Multiple testing correction怎么进行category wide tests of taxa的。
根据审稿人的其他意见,如第二条是2. Revise your bioinformatic pipeline as it is incorrectly stated as it stands for the data displayed. ;都是针对高通量测序数据的信息处理中的,都是比较偏重技术性的意见。
根据这些我初步推测,审稿人是不是要求用Multiple testing correction检验OTU系统分类的正确性的?这些想请各位大神指点迷津。另外,如果是针对OTU分类正确性的多重检验,那应该用什么软件进行处理?

2. Revise your bioinformatic pipeline as it is incorrectly stated as it stands for the data displayed.
3. Provide a percent failure for sequence reads that did not cluster into closed reference OTU picking in your PICRUSt analysis.
4. Revise to include the PICRUSt analysis in your results section.
