1.localize the gene on the genome, copy DNA sequence/region from the downstream of the former gene to closely upstream of your target gene (around 100-1000 bp).
2. paste the sequence into an online software for prediction of promoter core region, like BPROM.(http://linux1.softberry.com/berr ... mp;subgroup=gfindb)
3. Clone regions containing the core promoter if positive.
PS, if your target gene is one component of an operon, copy sequence upstream of the first gene of this operon for prediction and cloning. |
