[求助] PCR扩增出现的问题

我从一个8K质粒上扩增一段2.8K的目的片段,无论自己设计引物(16到22个碱基)还是按文献提供的引物(35和38个碱基),均扩增不出来。已经花了一个月的时间了,如果再扩增不出来,下个月就要换课题了。
目的片度的特点是GC含量约30%。
各位虫友,有什么好的建议吗?对我来说很重要,就麻烦大家了。

设计引物扩增目的片段跟目的片段的GC含量有什么关系,只要引物设计没有什么问题我觉得应该没有什么扩增不出来的条带吧?

你何不把你设计的引物发上来大家给你看看是不是有神马问题。

好吧,这么说吧:
1。确保你的模板没有任何问题,质粒用试剂盒重新抽提。
2.你要扩增的片段是从640-3490,AT含量较高,引物不好设计,你应该考试试专门针对AT-rich的扩增方法。
3.如果还是不行的话,建议你不要再纠结于引物扩增,而改用酶切回收,看了一下酶切位点还是有很多可以选择的,提供两个和你两端比较接近的
5‘端: BamHI 2   G/GATCC        635     5578
3'端:SwaI  2   ATTT/AAAT        3470    6123
直接回收635-3470片段大小的,
后面看你想干嘛了,用Taq酶补平加A尾进行TA克隆;或者再用你的引物PCR试试,应该没有什么太麻烦的二级结构了
