[求助] 挑菌测序,整成这样婶儿的结果,也是跪 已有1人参与

用植物材料提取的RNA、逆转录后作为模板PCR,之后连T载体挑菌,目标条带是1000bp,菌液PCR后只有一个接上目标条带,其他有数个连进了600bp的,想知道是什么,就一起送生工测序了。

测序结果:1000多的是目标片段,600多的中间缺400bp,其他部位和目标条带一毛一样。DNAMAN Alignment得到的。

我的想法:不可能是内含子吧,我是RNA逆转录的,引物也是按照转录组拼接得到的序列设计的。

可能1.600多的是植物自身的,是另一个基因?
可能2.产物断裂后,自连缺失了400bp,再接入T载体?

各位怎么想?求教!!!

Many thanks for you kind advice. I'll try to isolate genomic DNA as template.

What is the size of your amplification if the template uses the genomic DNA?

According the results your described above, I doubt that the approximate 1000 bp fragment came from genomic DNA contaminant. Therefore, the 600 bp fragment would be the target DNA.

我提取的RNA未进行去除基因组DNA操作,直接逆转录的,但是我是依照转录组拼接序列设计引物的,目标条带1000bp,应该不存在你说的问题。。

Can you confirm that your primers designed were cross-intron? Therefore, you should use the genomic DNA as the template for a positive test.
