请问虫子们:你们有没有做过油脂里的总酚含量测定?我们现在测遇到了一些问题,很无助,周围都没有人在做,不知道该怎么继续了?

请问你用的什么方法,福林酚法测的吗,出现什么问题了?个人觉得要测的话你得先把总酚提取出来再测,我只测过果汁的,用的是福林酚法和高效液相

是的,福林酚法,我们是测油的总酚含量,先用正己烷和甲醇提取出来,再测,按照文献上的操作,但是吸光度却很低,假若增加样品溶液的量又会产生沉淀。Briefly for the extraction of phenolic compounds,
10 g of oil was mixed with hexane (50 ml). This mixture
was placed into a separation funnel and 20 ml of methanol:
water (60:40, w:w) was added. The mixture was
shaken for 2 min and the separated water phase was
removed. This procedure was applied three times and all
the water phases were combined and used for TP analysis.
For TP analysis, the mixture of the sample solution
(50 ll), distilled water (3 ml), 250 ll Folin–Ciocalteu’s
reagent solution, and 7% Na2CO3 (750 ll) was vortexed
and incubated for 8 min at room temperature. Then
950 ll of distilled water was added. The mixture was
allowed to stand for 2 h at room temperature. The
absorbance was measured at 765 nm against distilled
water as blank

吸光值低,可能是总酚含量太低,可以考虑浓缩一下提取液,用旋转蒸发浓缩,或者真空干燥,这样试试看。要么就换个提取方法。
