[求助] 细胞DNA的提取已有2人参与

各位大家好，最近用QIAgen的试剂盒提取细胞的DNA，结果发现OD260/280数值为2.04左右，电泳图也显示有RNA污染，于是200ul体积加入4ul（100mg/ml）的RNA酶（技术推荐的加入体积），作用了10分钟，可是结果还是和以前的一样

，不知道该怎么办，都过去4个月了还是没有解决，网上说RNA酶作用15分钟，请各位知道的帮帮忙，谢了

The protocol above is used to treat RAN contaminants.

If you want to separate DNA from tissue cells, you should follow the protocol below.

1. Add 500 ul of pre-warmed 3% CTAB [100mM Tris-HCl, pH8.0, 25mM EDTA, 1.5M NaCl, 3%(W/V) CTAB, 1%(W/V) PVP] + 5 ul of 2-mercaptoethanol.
2. Incubate at 65 degrees Celsiu for 30 minutes.
3. Let the tubes cool down a little bit.
4. Add 500 ul of chloroform-isoamylalcohol 24:1, vortex.
5. Spin at full speed for 15 minutes.
6. Move the aqueous phase to a new tube.
7. Add 250 ul of 5M NaCl, mix.
8. Add 500 ul of cold isopropanol, mix.
9. Leave at least 20 minutes (up to overnight) at -20 degrees Celsiu.
10. Spin at max speed for 15 minutes.
11. Wash pellet with 1 ml of cold 75% ethanol.
12. Spin at max speed for 5 minutes.
13. Repeat wash.
14. Dry pellet.
15. Add 400 ul of high salt TE (1M NaCl in TE buffer) +2 ul of RNaseA (10mg/mL)
16. Incubate at 37 degrees Celsiu for one hour, mixing from time to time until the pellet dissolves
17. add 2 volume of cold 100% ethanol, put at -20 degree Celsiu for 20 minutes
18. Spin at max speed for 15 minutes
19. Wash pellet with 1ml of cold 75% ethanol
20. Spin at max speed for 5 minutes
21. Repeat wash
22. Dry pellet and resuspend in 100 ul of TE (or more, as you prefer)
