[交流] 【求助/交流】我的蛋白哪去了？焦虑啊！已有9人参与

各位大侠，能否帮我分析一下，我昨天刚刚提取的蛋白，当时就加buffer煮沸5min,然后跑电泳,考马斯亮蓝染色，效果很好,蛋白条带很清晰，今天想转膜，所以每个样品都拿出来煮了4min，跑两个泳道，一个做考马斯亮蓝染色，另一个转膜（其余操作和昨天一样）。但是结果不如人意，和前面的一样，做考马斯亮蓝染色后，除了marker以外，没有其他条带，转膜的结果也是这样，不知道我的蛋白哪去了？大家能否帮我分析一下原因。非常感谢。

Description:
Crude cell extracts contain a number of endogenous enzymes, such as proteases and phosphatases, which are capable of degrading the proteins present in the extract. The best way to improve the yield of intact proteins is to add inhibitors of  these enzymes known to be present in the source material. This series of protease inhibitor cocktail contain a mixture of protease inhibitors with a broad specificity for the inhibition of serine, cysteine, aspartic, metalloproteases, and  aminopeptidases etc. which have been optimized and tested for bacterial, fungal /yeast, plant and mammalian cell proteinextraction.
Applications:One vial of cocktail is recommend for inhibition of 20g tissues or 109cells in general (BS381 for 4g bacteria).
