最近投了一篇关于基因治疗方面的文章，下面是审稿人给出的意见，个人感觉挺抽象的，不知道大家有没有什么好的建议？特别是该如何回答呢？谢谢。 Reviewer #1: The results of the authors demonstrated that rAAV2-mediated β-globin virus can efficiently mediate the expression of normal β-globin gene in fetal hematopoietic cells from β-thalassemia patients, for at least 21-35 days in the recipient mice and the level of human β-globin expression significantly increased by 1.2-1.8 folds, as compared with that of controls. So, for the final potential use of rAAV-based clinical gene therapy for human β-thalassemia, more experimental works should be done to answer the questions: ⑴Actually how are the durability and the stability of the rAAV-based transduction? ⑵How is the level of peripheral hemoglobin after the rAAV-based gene therapy? |
