Measurement of H2S content H2S quantification was performed as described by Nashef et al. (1977). The chemical treatments were the same as the measurements of Na+ and K+ contents. Following the treatments, the root of seedlings were ground into fine powder with a mortar and pestle under liquid nitrogen and 0.3 g of frozen tissue were homogenized in 1 ml 100 mM potassium phosphate buffer (pH 7) containing 10 mM ethylenediaminetetraacetic acid (EDTA). The homogenate was centrifuged at 15,000¡Ág for 20 min at 4 ¡ãC, and 100 ¦Ìl of the supernatant was used for the quantification of H2S, in an assay mixture also containing 1,880 ¦Ìl extraction buffer and 20 ¦Ìl 20 mM 5,5¡ä- dithiobis (2-nitrobenzoic acid), to a total volume of 2 ml. The assay mixture was incubated at room temperature for 2 min and the absorbance was read at 412 nm.
