请教质谱（3Q）定量时，定量离子的选择问题 已有1人参与

本人投了一篇3Q定量的文章，定量方式是：在负离子模式下，采用二级质谱的子离子进行定量。但由于在流动相中加入了甲酸铵（ammonium formate），产生了[M+45]-和[M-H]-2个母离子（[M-H]的强度大于[M+45]），我是以[M-H]-母离子裂解后的子离子[A]-进行定量的。审稿人返回来的意见，认为[M+45]-也应该计算在含量中，给出一个相对值。 审稿人意见如下： I still would prefer a bit more explanation from the authors regarding my comment when ammonium formate was used as mobile phase additive, because as the authors mentioned, there will be [M+45]- adduct present together with [M-H]-, although the relative intensity changed as FV value increased. Considering different MS signal response for different analytes, should the intensity of this [M+45]- adduct also be counted, together with the intensity contribution from [M-H]-, to quantify the absolute total amount of a specific analytes? Would it make sense to add a column in Table 5 of the relative quantitative analysis rather than absolute quantitation (i.e., % of each individual triterpene acids in FLP and BFL, respectively), so that the difference of FLP and BFL is more evident to read? 对于审稿人的意见，我的理解是否有误？ 请各位大侠们，帮我看看。问题该如何处理，该如何回答？ |
