Gel filtration chromatography separates proteins in order of large to small molecules. In general, large proteins will not enter the gel pores and will elute rapidly from the column in the void volume. Smaller proteins will repeatedly enter and leave the pores of the gel and therefore will remain longer in the column. Hence, proteins will elute in order of decreasing molecular size.
