The enantiomericpuritydeterminationofasyntheticintermediateofnew3,4-dihydro-2,2-dimethyl-2H- 1-benzopyrans, i.e.4-amino-2,2-dimethyl-6-ethoxycarbonylamino-3,4-dihydro-2H-1-benzopyran, was successfully carriedoutusingananioniccyclodextrin(CD)derivativecombinedwithachiralionic liquid (IL).Inordertoobtainhighresolutionandefficiencyvalues,theadditionofachiralIL,i.e. ethylcholine bis(trifluoromethylsulfonyl)imide(EtCholNTf2), tothebackgroundelectrolytecontain- ing heptakis(2,3-di-O-methyl-6-O-sulfo)--CD (HDMS--CD) wasfoundtobeessential.Asimultaneous increase inseparationselectivityandenantioresolutionseemstoindicateasynergisticeffectofHDMS-- CD andEtCholNTf2. Thebestenantioseparationofthekeyintermediatewasachievedusingamethanolic solution of0.75Mformicacid,10mMammoniumformate,1.5mMHDMS--CD and5mMEtCholNTf2. Levamisole wasselectedasinternalstandard.Theoptimizedconditionsallowedthedeterminationof 0.1% ofeachenantiomerinthepresenceofitsstereoisomerusingthemethodofstandardadditions.The NACE methodwasthenfullyvalidatedwithrespecttoselectivity,responsefunction,trueness,precision,accuracy, linearityandlimitsofdetectionandquantification. |

本文建立了新的合成中间体3,4-二氢-2,2-二甲基-2H- 1-苯吡喃, 等.4-氨基-2,2-二甲基-6-乙氧基氨基-3,4-二氢-2H-1-苯并吡喃,的对映体纯度检查方法。 该方法使用阴离子环糊精结合手性离子液体衍生化。为了获得有效的分离和高分离度，实验中加入了手性离子液体，乙基胆碱双丙烯酰胺二酰亚胺，最重要的是背景电解质含有7-2，3-氧甲基-6氧-磺基 白塔环糊精，阴离子环糊精和手性离子液体的协同作用使得分离度和手性选择性同时增加。 这个重要的中间产物的手性分离条件是：流动相：含0.75M甲酸的甲醇溶液，10mM的醋酸铵缓冲溶液，1.5mM的阴离子环糊精和5mM的手性离子液体。 选用左旋咪唑作为内标物质。本方法在最佳的检测条件下可以检测到含有0.1%左旋或者右旋对映体杂质的样品。NACE检测和定量方法做了全面的方法学验证，包括选择性，精密度，准确度，线性范围， |

The enantiomeric purity determination of a synthetic intermediate of new 3,4-dihydro-2,2-dimethyl-2H- 1-benzopyrans, i.e.4-amino-2,2-dimethyl-6-ethoxycarbonylamino-3,4-dihydro-2H-1-benzopyran, was successfully carried out using an anionic cyclodextrin(CD) derivative combined with a chiral ionic liquid (IL).In order to obtain high resolution an defficiency values,the addition of a chiralIL,i.e. ethylcholine bis(trifluoromethylsulfonyl)imide(EtCholNTf2), to the background electrolytecontain- ing heptakis(2,3-di-O-methyl-6-O-sulfo)--CD (HDMS--CD) was found to be essential.A simultaneous increase in separation selectivity andenantioresolution seems to indicate asynergistic effect of HDMS-- CD and EtCholNTf2. The best enantioseparation of the key intermediate was achieved using a methanolic solution of0.75M formic acid,10mM ammonium formate,1.5mMHDMS--CD and 5mM EtCholNTf2. Levamisole was selected as internal standard.The optimized conditions allowed the determination of 0.1% of each enantiomer in the presence of its stereoisomer using the method of standard additions.The NACE method was then fully validated with respect to selectivity,response function,trueness,precision,accuracy, linearity and limits of detection and quantification. |
