http://xdrug.dxy.cn/bbs/topic/32884391 5楼说到核心问题了，这个问题云无心与方舟子PK过。方舟子曾说过“蛋白质经口服到了肠道，将会被消化成氨基酸才被人体吸收，通常不能直接进入人体发挥其独特作用，所以蛋白质药物(例如胰岛素)要注射才有效，口服则无效 ”，注意有“通常”二字。我正是要找自然界中极为少数的能被人肠道吸收的酶（但不是胰岛素等人内源蛋白），从物种种类方面估算，找到菌株的概率为1%，从酶的多样性上估算，找到目标酶的概率为1‰。请看云无心的科普文章：蛋白质进肚全成氨基酸？让实验说话；http://songshuhui.net/archives/43522 ；参考文献自己找吧。 |

http://xdrug.dxy.cn/bbs/topic/32884391 问题解析： 1、首过效应：最优为耐酸、抗胃蛋白酶、胰蛋白酶等，但也可采用包埋处理的肠溶剂型，减少首过损耗； 2、吸收效率(生物利用度)：大分子酶进入肠淋巴以及血循环的效率取决于酶特性，通常武功强于胃、胰蛋白酶、血浆蛋白酶的为优选，但还需试验、测试和筛选； 3、血浆半衰期：抗血浆中的各种蛋白酶，半衰期较长； 4、溶栓特性：就如清洗医用器械（如内窥镜官腔）的多酶清洗剂及自动循环清洗机。优选能直接溶栓但不激活内源纤溶系统，对形成周期长且半衰期长的胶原蛋白、纤维蛋白等能逐渐水解（靶点是血和淋巴循环中细胞外的各种蛋白，各种蛋白的生成和降解是动态的，半衰期20秒到1000天不等，如胶原蛋白的半衰期为1000天），在内源蛋白降解和生成的安全平衡区间内，用药疗程在3-6个月。酶的化学反应速度远快过血液流速，一般不考虑局部用药； 5、活性高、特异性高、安全性高、治愈率高、作用机理明确将是其主要特点。 （关于口服胰岛素的乱弹：胰岛素有51个氨基酸，一般肠吸收酶有500个左右氨基酸，融合不行，整合能行吗？即使能整合，酶及肠吸收特性又如何？自身会降解成那些组分？这个技术难度极大和未知数太多！） |
