293-F and 293-H Cells are clonal isolates, derived from transformed embryonal human kidney cells (1,2). The 293-F strain is a fast-growing variant of the 293 cell line. The 293-H strain is a variant of the 293 cell line that demonstrates better adherence in monolayer culture and ease of use for plaque assays and other anchorage dependent applications. The 293T cells are derived from 293 cells but stably express the SV40 large T antigen which can bind to SV40 enhancers of expression vectors to increase protein production. 293T are also neomycin resistant due to the presence of a neomycin resistance cassete together with the SV40 Large T . In addition to the comments above, the presence of T-antigen in 293T cells could help in episomal maintenance of SV40 origin containing vectors. In case of viral production and for other transfections 293T is now preferred since they are normally better transfectable. |
