[求助] 求助两道基因方面的问题,跪求大神解答。。。

1.

The white (w) gene is on the X chromosome and its loss of function causes loss of the red eye color in flies. However, the White protein is also expressed in the brain. You are curious what the function of w  in the brain might be, and you suspect its loss may cause neurodegeneration. You have access to multiple w alleles and you may use any reagents you can find in the literature.

Design two different genetic experiments to test the effect of a complete loss of function of w on neuronal health and survival in adult brains.

A hypothetical gene consist out of 6 exons. The translation initiation and stop codon are localized in exon 1 and 6, respectively. The gene is only expressed in adult mice in neurons in the brain and the heart muscle cells. Due to alternative splicing two different transcripts are found in the brain one with and one without exon 5 sequences. In the heart only a transcript without exon 5 sequences is found. The significance of the brain-specific transcript is not understood. Ablation of the transcript including exon 5 sequences could be instrumental to unravel its significance.

a)        Design a strategy including targeting construct for homologous recombination to ablate only the expression of the transcript including the exon 5 sequences.

b)        Which of the following Cre-transgenic mice could be of use in course of this research project: a neuron-specific Cre-transgenic mouse, a heart-muscle-specific Cre-transgenic mouse, a general deleter Cre- transgenic mouse or a Cre-transgenic mouse, in which Cre expression can be turned on in all cells by administration of tetracycline? Please explain for every Cre-mouse whether it could be of use.

c)        Could some kind of anti-sense strategy be an alternative approach? Explain your answer.
