| 细胞膜上有抗去垢剂区域吗？detergent-resistant membranes，有没有大神懂得给解释一下，抗去垢剂膜和病毒内吞有何联系？因为一篇文章中说病毒的一些非结构蛋白会和抗去垢剂膜相联系，感谢。。。 |

History • 80’s Order (package) and disorder phases in diff. parts of the PM. • 90’s Analysis of membrane fractions by the use of mild detergents. The concept of Detergent Resistant Membranes (DRM) born. In this technique non-solubilized fraction of the PM extracts by mild detergent (triton X-100 at 4ºC –artifact conditions-) + sucrose gradient • 2000’s New microscopy techniques: FRET, STED, FRAP… To understand the dynamic of the assembly of sterol, sphingolipds and proteins in living cells (real conditions). Membrane domain definition: Lateral compartment (PM plane) with composition, structure and biological function diff. from the surrounding membrane. Regardless size, stability and mechanism of formation. Why DRMs/DIMs (detergent insoluble microdomains) are not lipid rafts??? • 4ºC is not a real condition for the behavior of the PM. • Quantitative bias due the ratio concentration of detergent:prot. /detergent:lipids Definition by consensus 2006: “Membrane rafts are small (10-200nm), heterogeneous highly dynamic, sterol and sphingolipid-enriched domains that compartmentalized cellular process” What is new here: No: detergent resistant, lipid term, or PM (E.g. membrane of mitochondria), small (need of superesolution techniques). |
