The process for producing antibiotic composition Platenomycin by bioconversion of Midecamycin by A was investigated. The highest yield of Platenomycin was 5.654g/L. Recovering the antibiotic from the fermentation broth by adjusting the pH, and purifying the crude residue to 97.74%. The purified compound was identified as Platenomycin by MALDI-TOF-MS and NMR. Results demonstrated that this macrolide is active against Staphyloccocus aureu¡¢Streptococcus pneumoniae¡¢Septic Streptococcus and Porcine Actinobacillus pleuropneumoniae. | Platenomycin is a kind of macrolide antibiotic. By using A,we established a method to turn Midecamycin into Pratt neomycin ,with which fermentation production come to 5.528g / L. Through adjusting the PH to repeatedly precipitation and dissolve the crude extract combined with ODS reversed phase chromatography method ,the Pratt ADM A1 in the fermentation broth were isolated and purified.The product is of 97.74% purity and identified of Pratt neomycin by MALDI-TOF-MS and NMR detection. In vitro efficacy trials conducted of the purified Pratt ADM A1 verified that Pratt ADM has better drug sensitivity on Staphyloccocus aureu,Streptococcus pneumoniae,Septic Streptococcus and Porcine Actinobacillus pleuropneumoniae. |
