Ten microliters of protein (0.25 mgÆml)1) and 10 ll of saffron leaf total RNA (10 lgÆll)1) were mixed with 100 ll 50 mm MES (pH 6.0) and incubated at 50 C for 30 min. The reaction was stopped by adding 1 ml of ice-cold 4 m LiCl to the mixture, and incubated in ice for 15 min. Insoluble leaf total RNA was removed by centrifugation (10,000 g) for 15 min at 4 C. The supernatant containing soluble leaf total RNA was quantified by UV absorbance at 260 nm in a spectrophotometer (Lan Optics). One unit of enzyme was defined as the amount of protein causing an increase of absorption of 1.0 after 30 min of incubation.
