[求助] assay of ribonuclease activity

Ten microliters of protein (0.25 mgÆml)1) and 10 ll of saffron leaf total RNA (10 lgÆll)1) were mixed with 100 ll 50 mm MES (pH 6.0) and incubated at 50 C for 30 min. The reaction was stopped by adding 1 ml of ice-cold 4 m LiCl to the mixture, and incubated in ice for 15 min. Insoluble leaf total RNA was removed by centrifugation (10,000 g) for 15 min at 4 C. The supernatant containing soluble leaf total RNA was quantified by UV absorbance at 260 nm in a spectrophotometer (Lan Optics). One unit of enzyme was defined as the amount of protein causing an increase of absorption of 1.0 after 30 min of incubation. The positive and negative controls were RNase A and the heated CsPR10 (100 C for 10 min), respectively.

ribonuclease是RNA酶，也就是通常我们说的RNase。实验非常简单，是在RNA中加入RNase测定RNase的活性。然后通过测紫外吸收，如果RNA被降解，吸收减小。
