Determination of free amine groups [37]. ¼ì²â°±»ùµÄ
Dried gelatin samples (2¨C4 mg)
were reacted with TNBS (0.01 M) in a sodium hydrogencarbonate solution (pH
8.2, 4 w/v%, 2 ml) for 2 h at 40¡ÀC. Then hydrochloric acid (6 M, 3 ml) was
added to hydrolyse the gelatin gels at 60¡ÀC for 1.5 h. After cooling to room
temperature, deionised water (5 ml) was added to the solution and the absorbance
was measured against a TNBS-solution without gelatin, which had been treated
exactly the same as the other samples, at ¸ 345 nmwith an Uvikon 930 spectrometer.
The amount of free amine groups per 1000 amino acids was calculated using the
absorption coefŽ cient for 2,4,6-trinitrophenyl derivatised (hydroxy)lysine residues
(" D 14 600 lmol¡1 cm¡1 ), and assuming a molecular weight of 105 g mol¡1 for a
gelatin molecular chain of 1000 amino acids. The experiments were performed in
triplicate.

Titration of free carboxylic acid groups. ¼ì²âôÈ»ùµÄ
The titration of the cross-linked gels
was carried out using a protein titration procedure according to Kenchington et al.
[38, 39]. This procedure was adapted for titration of small volumes of insoluble
materials. The gelatin gels were swollen in deionised water for 1 h, powdered and
lyophilised. Deionised water (1.5 ml) as such or containing powdered gelatin gel
(50 mg) at 40¡ÀC was either titrated with hydrochloric acid (0.2 M) until pH 1.5 or
with sodium hydroxide (0.2 M) until pH 12, by adding 0.005 ml per step, and an
equilibration time of 60 s per addition.
