ReceivedJuly14,2006;RevisedManuscriptReceivedNovember13,2006 Thispaperdescribesthebioconjugationofhistidine-taggedenzymesandotherproteinstothesurfaceofcomposite !àmagnetomicelles!–consistingofmagnet-Fe O nanoparticlesencapsulatedwithincross-linkedpolystyrene- 2 3 block-polyacrylatecopolymermicelleshells.Freecarboxylicacidgroupsonthemagnetomicellesurfacewere 2+ convertedtoCu -iminodiaceticacid(IDA)forproteincapture.TheconjugationofT4DNAligaseandenhanced greenfluorescentproteintomagnetomicellesrevealedthatproteinswerecapturedwithahighsurfacedensity andcouldbemagneticallyseparatedfromreactionmixturesandsubsequentlyreleasedfromthenanoparticle surface.Additionally,bioconjugationofT7RNApolymeraseyieldedafunctionalenzymethatmaintainedits biologicalactivityandcouldberecycledforuptothreesubsequenttranscriptionreactions.Weproposethat protein-magnetomicellebioconjugatesareeffectiveforproteinbioseparationandenzymaticrecyclingandfurther strengthentheideathatnanoparticlesurfaceshaveutilityinproteinimmobilization. |
