Gels were formed by mixing the chosen PEG-acrylate with either dithiothreitol (DTT, Sigma) or PEG-dithiol, Mw ) 3.1 kD (Shearwater Polymers) at a 1:1 stoichiometric ratio of acrylates to thiols in 50 mM phosphate buffered solution (PBS, pH 7.8). Briefly, appropriate amounts of each reactant were dissolved separately in an aliquot of PBS. The amount of PBS was varied to give the desired total precursor concentration (wt %) upon mixing. The two solutions were mixed vigorously in 1.5 mL plastic tubes and centrifuged briefly to remove bubbles. The sealed tube containing the mixture was then placed at 37 °C, and the gel was allowed to react overnight to ensure complete conversion. For almost all systems, gelation was observed visually within a few minutes of mixing. 特别是其中的50mM，vigorously应该怎么理解？ |
