The preparation method, as far as one can tell, involves the addition of dried plasma components which are in the supernatant fraction after centrifugation. They add these components to the solution of the anaesthtic and the dye, at amounts which are not clearly mentioned. However, to claim that this method actually can detect these molecules in a patient’s blood plasma, one must add these components to the plasma before processing it in any way. Then and then only one would be able to know how much the background components in the plasma would interfere with the detection.

根据现有证据来看，准备方法包括加入存在于离心后上清中的干血浆成分。他们将这些成分加到麻醉剂和染料溶液中，分量不清楚。然而，确声称这种方法能检测病人血浆中的这些分子，不管怎样，得事先在血浆中加入这些成分。那么就只有天知道血浆中有多少背景成分干扰这个检测。

谢谢您的回答，我不是很明白到底这个审稿人要求回答什么。我是这样准备样品的:首先将血浆涡旋离心后去上清液，在上清液中加入乙腈和一定浓度的麻醉药。然后再涡旋离心，取上清液置于真空干燥箱中除去有机溶剂。最后用流动相重新溶解样品，进样。
