The effects of formic acid and acetic acid on the separation of DHT enantiomers were listed in Table 3. µ±Á÷¶¯ÏàÖв»Ìí¼Ó¼×ËáºÍÒÒËáʱ£¬DHTµÄÁ½¸ö¶ÔÓ³Ìå¿ÉÒÔ´ïµ½»ùÏß·ÖÀë¡£¼øÓÚÖù×ÓµÄÄÍÓÃÐÔ£¬Á÷¶¯ÏàÖÐËáÌí¼ÓÁ¿ÔÚ 0. 1% ~ 0.2% Ö®¼äʱ£¬ DHT¶ÔÓ³Ìå¿ÉͬʱʵÏÖ»ùÏß·ÖÀë¡£ Ϊ»ñµÃ½ÏºÃµÄ·ÖÀëЧ¹û£¬²¢¾¡Á¿½ÏÉÙ±£Áôʱ¼ä£¬×îÖÕÑ¡ÔñÌí¼Ó0.2%¼×Ëá¡£When the separation was conducted using a mobile phase without formic acid and acetic acid, a baseline separation could (not?) be achieved for the two enantiomers of DHT. DHT was a weak acidic material. Adding certain amount of acid (formic acid and acetic acid were used in this experiment) into mobile phase in HPLC analysis chould reduce tailed chromatographic peaks and increase its degree of symmetry. The effects of the additive amount of formic acid and acetic acid in mobile phase to DHT enantiomeric separation were listed in table 3. The two DHT enantiomorphs chould achieve baseline separation, without no acid(formic acid and acetic acid) addition in mobile phase. Considering the durability of column, DHT enantiomorphs were simultaneous achieved baseline separation when acid addition was 0. 1% ~ 0.2%. Furthermore, 0.2% formic acid addition was final chose to obtain better separation effect and less retention time simultaneously. |
