摘  要

本研究通过对鼠伤寒沙门菌SL1344及Δcrp缺失株对HeLa细胞黏附和侵入的检测,分析crp基因与黏附和侵入的相关性,为研究开发更安全有效的弱毒苗奠定基础。为了研究鼠伤寒沙门菌SL1344及Δcrp缺失株HeLa细胞黏附和侵入,对沙门菌进行活化、稀释涂板,过夜培养后计数;对HeLa细胞复苏、传代、冻存及计数,待细胞数目合适时移至24孔细胞板进行过夜培养。随后进行黏附试验,即在细菌接种前2h,将细胞培养基换为含10%FBS、无抗生素的DMEM培养基。取对数生长期的细菌(SL1344株及SL1344Δcrp突变菌),离心并用无菌PBS洗漆细菌,用无血清无抗生素的DMEM悬浮细菌,以MOI比值100:1加入细胞中。常温下离心,使细菌与细胞充分接触。孵育一定时间后,用无菌PBS洗涤细胞,接着用Triton X-100溶液裂解细胞。随后进行稀释,然后选取3个合适的稀释度涂布LB平板,各做3个重复,细菌计数。而侵入实验是在孵育和无菌PBS洗涤细胞3次后,用加入含10%FBS和高浓度庆大霉素的DMEM培养基培养细胞,其他同黏附实验的步骤。实验结果表明与SL1344亲本菌株相比,Δcrp缺失株对HeLa细胞的侵袭力与黏附率降低,表明具有一定的免疫原性,这为研制安全有效的鼠伤寒沙门菌疫苗以及疫苗载体奠定了基础。,

Abstract
本研究通过对鼠伤寒沙门菌SL1344及Δcrp缺失株对HeLa细胞黏附和侵入的检测,分析crp基因与黏附和侵入的相关性,为研究开发更安全有效的弱毒苗奠定基础。
The correlation between crp gene and  adhesion and invasion was analyzed in this study based on the detection of that salmonella typhimurium SL1344 and Δcrp deleted strain adhered and invaded the HeLa cell, which has established a foundation for developing more  safety and efficacy attenuated vaccine.
为了研究鼠伤寒沙门菌SL1344及Δcrp缺失株HeLa细胞黏附和侵入,对沙门菌进行活化、稀释涂板,过夜培养后计数;对HeLa细胞复苏、传代、冻存及计数,待细胞数目合适时移至24孔细胞板进行过夜培养.
For detecting the  adhesion and invasion, salmonella was activated and dilution painted, while the HeLa cell,which was  resuscitated, passaged,  cryopreserved and counted, was moved into 24-hole cell plate to  overnight culture after  suitable cell number.
随后进行黏附试验,即在细菌接种前2h,将细胞培养基换为含10%FBS 、无抗生素的DMEM培养基。
The  adhesion text was conducted later, namely that cell culture media was replaced by DMEM culture medium with 10%FBS and  without antibiotics.

取对数生长期的细菌(SL1344株及SL1344Δcrp突变菌),离心并用无菌PBS洗漆细菌,用无血清无抗生素的DMEM悬浮细菌,以MOI比值100:1加入细胞中。常温下离心,使细菌与细胞充分接触。
The bacteria(SL1344 strains andSL1344Δcrp mutant strain ) in the logarithmic phase were  selected to hatch by centrifugal method at room temperature to make sure complete contact between cell and  bacteria, which were leached by  sterility PBS and suspended  through the use of serum-free and  without antibiotics DMEM and finally added to the cell at MO  ratio was 100:1.
孵育一定时间后,用无菌PBS洗涤细胞,接着用Triton X-100溶液裂解细胞。随后进行稀释,然后选取3个合适的稀释度涂布LB平板,各做3个重复,细菌计数。
After certain time's  incubation, the cell was washed by  sterility PBS andfollowed was lysised by using Triton X-100 solution. Then, the lysate was diluted and 3 appropriate different dilution were select to coate the LB plate. The bacteria amount were calculated after 3 times coating experimental.
而侵入实验是在孵育和无菌PBS洗涤细胞3次后,用加入含10%FBS和高浓度庆大霉素的DMEM培养基培养细胞,其他同黏附实验的步骤。
While the cells were cultivated by using DMEM culture medium with 10% FBS and high concentration gentamicin after 3 times washed by the use of  sterility PBS in the process of  incubation in the thinvasion text, and the others  experimental steps were the same with the  adhesion text .
实验结果表明与SL1344亲本菌株相比,Δcrp缺失株对HeLa细胞的侵袭力与黏附率降低,表明具有一定的免疫原性,这为研制安全有效的鼠伤寒沙门菌疫苗以及疫苗载体奠定了基础。
Experiment result shows that compared with SL1344 parental strains, the ,the invasiveness and adhesive properties of the Δcrp deletion strains to HeLa cell were reduced, which indicating the cell has some immunogenicity. This study  established the base of   developing safety and efficacy  salmonella typhimurium  vaccine and vaccine vectors.
