摘 要
Abstract
本研究通过对鼠伤寒沙门菌SL1344及Δcrp缺失株对HeLa细胞黏附和侵入的检测,分析crp基因与黏附和侵入的相关性,为研究开发更安全有效的弱毒苗奠定基础。
The correlation between crp gene and adhesion and invasion was analyzed in this study based on the detection of that salmonella typhimurium SL1344 and Δcrp deleted strain adhered and invaded the HeLa cell, which has established a foundation for developing more safety and efficacy attenuated vaccine.
为了研究鼠伤寒沙门菌SL1344及Δcrp缺失株HeLa细胞黏附和侵入,对沙门菌进行活化、稀释涂板,过夜培养后计数;对HeLa细胞复苏、传代、冻存及计数,待细胞数目合适时移至24孔细胞板进行过夜培养.
For detecting the adhesion and invasion, salmonella was activated and dilution painted, while the HeLa cell,which was resuscitated, passaged, cryopreserved and counted, was moved into 24-hole cell plate to overnight culture after suitable cell number.
随后进行黏附试验,即在细菌接种前2h,将细胞培养基换为含10%FBS 、无抗生素的DMEM培养基。
The adhesion text was conducted later, namely that cell culture media was replaced by DMEM culture medium with 10%FBS and without antibiotics. |
