In this study, we used a two-step PCR protocol as previously described (19). With this approach, tags and adapters were added in a second round of PCR amplification. 额额，不好意思，The first PCR amplification was performed using the modified primers 341F (CCTAYGGGRBGCASCAG) and 806R (GGACTACNNGGGTATCTAAT) (20) to amplify a 466-bp fragment of the 16S rRNA gene flanking the V3 and V4 regions. The PCR mix (40 l) contained 1Phusion HF buffer containing 2.5 mM MgCl2 (Finnzymes, Vantaa, Finland),0.2mMdeoxynucleoside triphosphate (dNTP) mixture, 0.8UPhusion hot start DNA polymerase, 2 M each primer, 25 g bovine serum albumin (BSA; molecular grade, 10 mg ml1 stock), and 1 l template.先是扩增的16S rRNA，然后用这个DNA进行的第二轮扩增，应该就是RT-PCR了吧。。。 |
