Experiments of infrared spectroscopy, water absorption of weighing, the rotational rheometer, and MTT were conducted to measure the structure, swell ability, elastic modulus and cytotoxicity of polyacrylamide/chitosan hydrogel HepG2 cells were inoculated into the mixed hydrogel base material of polyacrylamide/chitosan, and the morphology of the cells grown on the materials was observed after 24h. The hardness of the different formulation of polyacrylamide/chitosan hydrogel had obvious gradient differences and good biocompatibility. This new type of the biological substrate material could contribute to the research of cell mechanics.
