Abstract: A new type of hardness variable biological substrate material was prepared. Experiments of infrared spectroscopy, water absorption of weighing, the rotational rheometer, and MTT were conducted to measure the structure, swell ability, elastic modulus and cytotoxicity of polyacrylamide/chitosan hydrogel HepG2 cells were inoculated into the mixed hydrogel base material of polyacrylamide/chitosan, and the morphology of the cells grown on the materials was observed after 24h. The hardness of the different formulation of polyacrylamide/chitosan hydrogel had obvious gradient differences and good biocompatibility. This new type of the biological substrate material could contribute to the research of cell mechanics.
