Suppression of Wnt signaling by the miR-29 family is mediated by demethylation of WIF-1 in non-small-cell lung cancer Wnt inhibitory factor-1 (WIF-1) silencing induced by promoter hypermethylation is a common mechanism of aberrant activation of the Wnt signaling pathway in non-small-cell lung cancer (NSCLC). However, the activity of regulators associated with the methylation of the WIF-1 gene remains unclear. Here, we investigated the role of three DNA methyltransferases (DNMT1, DNMT3A and DNMT3B) in the expression of WIF-1. The three DNMTs were up-regulated in NSCLC tumor tissues and suppression of DNMT3A and DNMT3B restored the expression of WIF-1 in NSCLC cells. The miR-29 family (miR-29a, -29b, and -29c), which negatively regulates DNMT3A and DNMT3B, was examined in association with the Wnt/b-catenin signaling pathway. A positive correlation between the expression of WIF-1 and that of MiR-29s was observed in NSCLC tissues. Methylation-specific PCR and Western blotting indicated that miR-29s positively regulate WIF-1 expression by inhibiting the methylation of its promoter. Furthermore, miR-29 overexpression downregulatedb-catenin expression, inhibited cell proliferation and induced apoptosis. The expression of miR-29a and miR-29b was partially regulated by DNMT3A and DNMT3B in a positive feedback loop. Taken together, our findings show that miR-29s suppress the Wnt signaling pathway through demethylation of WIF-1 in NSCLC |

在非小细胞肺癌，miR-29家族成员通过介导WIF-1去甲基化而抑制Wnt信号途径 通过启动子超甲基化使Wnt抑制因子－1（WIF-1）静默在非小细胞肺癌（NSCLC）中是引起Wnt信号通路异常激活的一种常见机制。但是，与WIF-1基因甲基化相关的调节因子的活性尚不清楚。 在此，我们研究了三种DNA甲基转移酶(DNMT1, DNMT3A and DNMT3B)在WIF-1表达中的作用。在NSCLC组织中这三种DNMTs的表达被向上调节，在NSCLC细胞中抑制DNMT3A和DNMT3B的表达可以恢复WIF-1的表达。miR-29家族 (miR-29a, -29b, and -29c)对DNMT3A和DNMT3B有负调节作用 ，我们研究了miR-29家族与Wnt/β-catenin信号通路之间的相互关系。结果发现，在NSCLC组织，WIF-1表达与miR-29s之间存在正相关。甲基化特异性PCR（聚合酶链式反应）和Western blotting（这个就不用翻译了吧？）表明miR-29s可以通过抑制WIF-1启动子的甲基化而正向调节其表达。 而且，miR29过表达可以下调β-catenin的表达，抑制细胞增值并诱导细胞凋亡。miR-29a和miR-29b的表达则在一正反馈回路中受DNMT3A和DNMT3B的部分调节。总之，我们的发现表明，在NSCLC miR-29s通过引起WIF-1去甲基化而抑制Wnt信号途径。 注：本人发表过多篇Wnt/β-catenin的相关文章，却从不知β-catenin该如何翻成中文，刚才查了下，好像有人称之为β－连环素，听上去很不习惯，也不知在说什么，所以我建议不用翻成中文了。 |
