叉头转录因子O亚家族1(forkhead transcription factor group O1,FoxO1)是前体脂肪细胞分化过程的重要调控因子,通过多条通路影响细胞增殖、分化、凋亡但其作用机制及信号通路在鱼类的研究中尚不清楚。本项目拟以我国重要经济鱼类一草鱼为实验材料,采用细胞培养、RT-PCR、实时定量PCR、载体构建、免疫组化、细胞转染、Western blotting及免疫共沉淀结合质谱鉴定技术等技术手段,以草鱼脂肪细胞为体外模型,以FoxO1调控转录因子PPARγ的活性、RES和Wortmannin阻断剂对FoxO1的影响及磷酸化FoxO1的亚细胞定位和作用为切入点,研究FoxO1基因对草鱼脂肪细胞增殖分化的调控作用及信号通路。研究结果不仅能为阐明脂质沉积的复杂网络调节机制奠定基础,而且还可改为善鱼肉质量与风味提供依据,具有重强的理论及应用意义。 [ Last edited by canfeng2008 on 2013-3-7 at 08:10 ] |

Great priorities of the research were given to the control of the activity of transcription actor PPARγ by FoxO1,the influence of RES and Wortmannin blockers on FoxO1 and the locallization and function of phosphorylated FoxO1 subcell ,in order to investigate on the modulation and signalpathways of the proliferation and differentiation of grass carp cell by FoxO1. 研究结果不仅能为阐明脂质沉积的复杂网络调节机制奠定基础,而且还可改为善鱼肉质量与风味提供依据,具有重强的理论及应用意义。 翻译:The research result will not only lay the foundation for the clarification of the regulatory mechanism of the complex network of lipid storage,but also provide the basis for improving the flavor and quality of fish,and it is of great significantce theoretically and practically |
