叉头转录因子O亚家族1（forkhead transcription factor group O1，FoxO1）是前体脂肪细胞分化过程的重要调控因子，通过多条通路影响细胞增殖、分化、凋亡但其作用机制及信号通路在鱼类的研究中尚不清楚。
翻译：FoxO1 is an important regulator in preadipocytes differentiation ,and it infulences the proliferation ,differentiation and apoptosis of cells through multiple pathways while the mechanism and signal pathways in the research of fish is not clear .
本项目拟以我国重要经济鱼类一草鱼为实验材料，采用细胞培养、RT-PCR、实时定量PCR、载体构建、免疫组化、细胞转染、Western blotting及免疫共沉淀结合质谱鉴定技术等技术手段，以草鱼脂肪细胞为体外模型，以FoxO1调控转录因子PPARγ的活性、RES和Wortmannin阻断剂对FoxO1的影响及磷酸化FoxO1的亚细胞定位和作用为切入点，研FoxO1基因对草鱼脂肪细胞增殖分化的调控作用及信号通路。
翻译：This project intends to choose grass carp，one of our impotant commercial fishes，as experimental materials.Cell culture , RT-PCR , real-time quantitative PCR , vector construction, immunohistochemistry , cell transfection , Western blotting and co-immunoprecipitation combined with mass spectrometric identification technology as the means.Adipocytes grass carp as the vitro model. Great priorities of the research were given to the control of the activity of transcription actor PPARγ by FoxO1，the influence of RES and Wortmannin blockers on FoxO1 and the locallization and function of phosphorylated FoxO1 subcell ，in order to investigate on the modulation and signalpathways of the proliferation and differentiation of grass carp cell by FoxO1.
