| Several groups have now clearly shown the anti-tumour effects of Dll4 blockade alone. γ-Secretase inhibitors also affect tumour growth through multiple mechanisms, as its targets include many substrates besides Notch cleavage.There has been dose-limiting gastrointestinal toxicity with those inhibitors, because of the key role of Notch signalling in differentiation of the gut. |

1）几个课题组现在已经清楚地表明Dll4阻塞剂本身就有抗肿瘤效应。 γ-分泌酶抑制剂也通过多种机理影响肿瘤的生长, 这是因为其靶向(物)除了Notch 裂解, 还包括许多物质。因为Notch信号在区分内脏中的关键作用，使用那些抑制剂时会有一定的胃肠道毒性，这种毒性和其使用的剂量有关。 以上理解的基础上，可以更进一步简化： 2）几个课题组现在已经清楚地表明，Dll4阻塞剂本身就有抗肿瘤效应。 γ-分泌酶抑制剂也是通过多种机理影响肿瘤的生长, 这是因为其靶向(物)除了Notch 裂解（位）, 还包括许多物质。因为Notch信号在区分内脏中的关键作用，因此那些抑制剂会有剂量限制的胃肠道毒性。 |
