D-maltose monohydrate (2.00 g, 5.55 mmol), (+)catechin hydrate (6.00 g, 20.67 mmol), and poly(ethylene glycol) dimethyl ether (Mn 500, 4.0 g) were mixed with 50 mM citrate buffer, pH 6.0 (19.8 mL), and heated to 95¡ãC to dissolve the reactants. The reaction mixture was transferred to a shaker-incubator (125 rpm) at 37¡ãC. After 20 min, a-D-glucosidase (100 U in 0.2 mL citric buffer) was added. The progress of the reaction was monitored by TLC as described above (Rfglucoside 4 0.48). After 24 h, the water was evaporated under vacuum 50¨C60¡ãC using Bu¡§chi rotor evaporator for about 1 h and the product was isolated on a silica gel column (25 ¡Á 2 cm) eluting with chloroform first, then using gradient elution with chloroform/methanol from 1:1 to 1:5 (v/v). The product (+)catechin maltoside (0.50 g, 1.11 mmol) was obtained in 20% yield, based on maltose. |
