流式细胞仪 测细胞凋亡率

请教用过流式细胞仪测细胞凋亡率的虫友，这里的凋亡率是根据什么来计算的？ 这里有一段文献是用Acridine orange/ethidium bromide (AO/EB) 染色的，内容如下：Then we confirmed FTFs induced apoptosis bymeans of acridine orange/ ethidium bromide double staining method. Acridineorange/ethidium bromide (AO/EB) staining was used to visualize nuclear changes and apoptotic body formation that were characteristic of apoptosis. The following four cellular states were recorded under the microfluoroscope: (i) viable cells colored green with normal nuclear structure (VN); (ii) early apoptotic cells stained green with condensed or broken nuclear (VA); (iii) late apoptotic cells that show orange color with condensed or broken nuclear (NVA); and (iv) necrotic cells that appeared orange with a normal nuclear structure (NVN). Apoptotic ratewas calculated according tothe number of different cell types (apoptotic rate = (VA + NVA)/ (VN + VA + NVA + NVN) *100%)。 我想问的是，PI染色的话，那这个apoptotic rate 的公式应该是怎样的呢？是不是也有这个颜色的区别啊？ 非常感谢！！！ |
