叶片中的激素提取不出来

Sample Text 我现在用下面的方法来提取草的叶片中的IAA，然后再用高效液相色谱测量，可是提取不出来，不知道有没有人用过这种方法，希望能指点我一下，不胜感激；或者有更好的方法。谢谢 （1）取0.5g叶片，加液氮研磨至粉碎，然后加入8ml（0.05M，PH=7.0，含0.02%二乙基二硫带氨基甲酸钠）磷酸钠缓冲液，继续研磨至匀浆，倒入离心管中， （2） 在4。C条件下持续振荡1~2h。 （3） 取出离心管，用1M盐酸调PH约2.6 （4） 加入0.5g XAD-7离子交换树脂，在4。C条件下振荡半小时。 |

Leaf tissue (20 mg) was ground with a mortar and a pestle in liquid N, and homogenized in 1.5 mL Na-phosphate buffer (0.05 M, pH 7.0) containing 0.02% (w/v) sodium diethyldithiocarbamate as an antioxidant. An internal standard of 13C6–IAA (200 ng) was added into each sample and extracted by continuous shaking for 1 h at 4°C. The samples were transferred into 1.5-mL microcentrifuge tubes after extraction and pH was adjusted to approximately 2.6 with 1.0 M HCl (about 43 μL of HCl was required for 1 mL of extract). The sample was slurried with 60 mg of Amberlite XAD-7 (Sigma, MO) for 30 min. After removal of the buff er, the XAD-7 was washed with 2 × 500 μL of 1% acetic acid before being slurried with 2 × 500 μL of dichloromethane for 2 × 30 min (Edlund et al., 1995). The combined dichloromethane fractions were reduced to dryness with nitrogen gas. Samples were methylated with ethereal diazomethane at room temperature according to Prinsen et al.
