[交流] 【求助/交流】求助一道中科院考博题

在高等生物蛋白组学研究中发现,逆境条件下某蛋白A含量远远高于它在正常条件下的含量,但是在mRNA水平上检测表明,在逆境条件和正常条件下此基因的表达水平基本一致。请根据你所学的知识说明其可能的表达调控机制。

one possibility is under stress conditions, the protein stability is much stronger than the normal conditions, means the half life is longer which might be due to post translationally modification

我觉得是在逆境的条件下,促进了该蛋白的翻译水平的提高,另外的可能是这种蛋白本身存在降解机制,但是由于逆境的出现,这种机制受到了抑制,导致该蛋白的积累。

you know the answer more than me
I know there are some genes which are not affected at mRNA level but the protein level is severely reduced, one reason is the specific mRNA can not be efficiently attached to the ribosome,

Just as you say, another reason is the effection of miRNA at translational level.  In this mode of action, miRNA usually targets the 3'-UTR of mRNA and inhibite the protein production.

in my study it is not the case you talked
there are some proteins which are involved in mRNA binding and target mRNA to ribosome, when these proteins are deleted, some specific mRNAs are still expressed but less mRNAs appear in the ribosome, therefore the protein synthesis is reduced

个人觉得应该是翻译水平的调控!但是也有可能是RNA编辑的结果

我有点不懂了了,楼主说的是蛋白质水平量表达升高,您这里说的是蛋白水平的降低,这是在说另外一个问题了吗?如果是蛋白质水平的降低的话,转录水平上没有变化的,还可以是转录后的水平,翻译水平以及翻译后的加工等都会影响蛋白质的表达,这里面似乎涉及非常多的因素,如果要真的回答这道题的话,应该从多个方面说吧。如果楼主问题中涉及到的蛋白是信号通路或者代谢途径中的问题的话,还有更多的要说。
