| 以NCBI发表的牛抗菌肽（pl-amp）为模板，在多肽C端添加adearein蛋白酶切位点串联起来，根据大肠杆菌的偏好密码子，反向翻译并合成设计后的成熟串联抗菌肽基，将合成的基因克隆入PET30a(+)载体中构建重组大肠杆菌表达载体PET30a(+)-pa,经酶切分析和PCR证明载体构建成功，PET30a(+)-pa的正确构建，为抗菌肽的来源问题奠定了基础。 |

| Using pl-amp as the template, adearein protease was added to the peptide C terminus in order to link cut sites together, according to the preferring codon of colibacillus, mature series antimicrobial peptides designed was backward translated and synthetized, then the synthetic gene was cloned into PET30a (+) vector to construct the recombinant colibacillus expression vector PET30a (+)-pa,by enzyme cutting analysis and PCR, the results showed that carrier construction succeeded, and the right construction of PET30a(+)-pa laid the foundation of the source problem for the antibacterial peptide. |
