Using pl-amp as the template, adearein protease was added to the peptide C terminus in order to link cut sites together, according to the preferring codon of colibacillus, mature series antimicrobial peptides designed was backward translated and synthetized, then the synthetic gene was cloned into PET30a (+) vector to construct the recombinant colibacillus expression vector PET30a (+)-pa,by enzyme cutting analysis and PCR, the results showed that carrier construction succeeded, and the right construction of PET30a(+)-pa laid the foundation of the source problem for the antibacterial peptide. |
