Amersham Pharmacia columns PD-10 are prepacked,disposable columns containing Sephadex® G-25 M for rapid desalting and buffer exchange. Dimensions Bed volume: 9.1 ml Bed height: 5 cm Eluent composition Columns PD-10 contain Sephadex G-25 M swollen indistilled water containing 0.15% Kathon® CG/ICP Biocide* as preservative. The columns can be rapidly equilibrated with other aqueous buffer solutions. Instructions for use 1. Remove the top cap and pour off the excess liquid. 2. Remove the bottom cap. 3. Support the column over a suitable receptacle and equilibrate the gel bed with approximately 25 ml of the required buffer solution. 4. When the equilibration buffer has run into the column, add the sample in a total volume of 2.5 ml. If the sample volume is less than 2.5 ml, add buffer until sample volume plus buffer volume 2.5 ml. Discard the eluent. 5. When the sample has run into the column, elute the high molecular weight components e.g. proteins, with 3.5 ml elution buffer solution. Note Small air bubbles along the plastic walls of the column and on the bottom membran may occur. This does not affect the performance of the column.
