The protein that was lost while washing the microspheres during the fabrication process was not accounted for when calculating the loading. Hence the loading and loading efficiency are slightly overestimated. The encapsulation efficiency of the protein was determined by dividing the mass of the loaded protein by the initial mass of protein. The entrapment could be determined in a reproducible and quantitative manner. |

In the synthesis procedure, the micocapsules were obtained by centrifuating the suspension, and the supernatant was also collected for further characterizations. Firstly, the amount of protein that was not encapsulated was known by the BSA content in the supernatant, which could be determined by means of BCA protein assay. Then, the amount of encapsulated BSA in the microspheres, therefore, could be easily calculated by mass balancing, during which the amount of protein that lost while leaching was not considered. On that account, the loading contents of the protein were overestimated with a very small deviation.
