PorousSiisananostructuredmaterialthatisofinterestformolecularandcell-basedbiosensing,drug livery,andtissueengineeringapplications.Surfacechemistryisanimportantfactordeterminingthe
stabilityofporousSiinaqueousmedia,itsafnityforvariousbiomolecularspecies,anditscompatibility
withtissues.Inthisstudy,theattachmentandviabilityofaprimarycelltypetoporousSisamples
containingvarioussurfacechemistriesisreported,andtheabilityoftheporousSilmstoretaintheir
opticalreectivitypropertiesrelevanttomolecularbiosensingisassessed.Fourchemicalspeciesgrafted

totheporousSisurfacearestudied:siliconoxide(viaozoneoxidation),dodecyl(viahydrosilylationwith

dodecene),undecanoicacid(viahydrosilylationwithundecylenicacid),andoligo(ethylene)glycol(via

hydrosilylationwithundecylenicacidfollowedbyanoligo(ethylene)glycolcouplingreaction).Fourier

TransformInfrared(FTIR)spectroscopyandcontactanglemeasurementsareusedtocharacterizethe

surface.Adhesionandshort-termviabilityofprimaryrathepatocytesonthesesurfaces,withand
withoutpre-adsorptionofcollagentypeI,areassessedusingvitaldyes(calcein-AMandethidium
homodimerI).Cellviabilityonundecanoicacid-terminatedporousSi,oxide-terminatedporousSi,and
oxide-terminatedat(non-porous)Siaremonitoredbyquanticationofalbuminproductionoverthe
courseof8days.ThestabilityofporousSithinlmsafter8daysincellcultureisprobedbymeasuring
theopticalinterferometricreectancespectra.Resultsshowthathepatocytesadherebettertosurfaces
coatedwithcollagen,andthatchemicalmodicationdoesnotexertadeleteriouseffectonprimaryrat
hepatocytes.ThehydrosilylationchemistrygreatlyimprovesthestabilityofporousSiincontactwith
culturedprimarycellswhileallowingcellcoveragelevelscomparabletostandardculturepreparations
ontissueculturepolystyrene
