PhD fellowship in peptide and protein delivery

Department of Pharmacy
Faculty of Health and Medical Sciences (SUND)
University of Copenhagen

Department of Pharmacy is offering a three-year PhD fellowship within the field of biopharmaceuticals and biophysics. The PhD fellowship is financed by the University of Copenhagen. The fellowship is available from 1 December 2016 or as soon as possible thereafter.

Project description
As a consequence of rapidly increasing resistance development in bacteria against conventional antibiotics, the interest for alternative therapeutics has increased. Antimicrobial peptides constitute one such class of potential therapeutics. In our earlier work, we have identified a battery of endogenous peptides displaying potent antimicrobial effects but simultaneously low toxicity against human cells. Some of these peptides also display potent anti-inflammatory effects, of considerable interest in both acute and chronic inflammation, as well as anti-cancer effects. However, efficient therapies based on such peptides, as well as other biomacromolecular drugs, also require efficient delivery systems, as such drugs are susceptible to both chemical and enzymatic degradation, and prone to conformational changes and/or aggregation, with pronounced consequences for functionality and safety.
The project aims at building new knowledge on peptide interactions with polymer-based microgels, and the use of the latter as peptide delivery systems, building on our previous work on factors affecting peptide and protein loading to, distribution within, and release from microgels, as well as biodegradation effects. In particular, focus in the project will be placed on employing physicochemical methodology for studies of effects on microgel properties (charge, size, cross-linking density, degradability) on their use as delivery systems for antimicrobial and related peptides, both in solution and at surfaces, the latter of interest for localized depot therapies and functional biomaterials. The work is performed using advanced biophysical methods, such as neutron scattering, ellipsometry/QCM, and confocal microscopy. Biophysical results are correlated to biological studies of cell uptake and toxicity, in collaboration with research groups specialized in such studies.
Principal supervisor
Professor Martin Malmsten, Department of Pharmacy (as of 1 October 2016)
