DNA isolation is a very error prone step.Significant different results might be obtained with different extraction methods and reagents. In addition, different researchers can also get dramatic different results with the same method and samples. It has been reported by many researchers that the recovery rate of DNA with different methods is quite different, in some cases, the difference is even up to several times. Moreover, most of the small DNA fragments will be lost when the DNA is purified with some commercial DNA isolation kits.
